摘    要:目的观察131I-GMCSF在诱导HL60/ADM细胞凋亡过程中细胞周期的改变,进一步探讨凋亡与细胞周期改变的关系.方法采用体外放射免疫治疗模型,通过流式细胞术观察131I-GMCSF诱导HL60/ADM细胞凋亡情况及其对HL60/ADM细胞周期的改变.结果 Annexin V/PI双染色法证实一定放射性浓度的131I-GMCSF能明显诱导HL60/ADM细胞凋亡,且随着作用时间的延长,细胞凋亡率增加,流式细胞术显示作用24h后的细胞周期改变主要表现为S期阻滞和G2/M期阻滞,而作用48h后则主要引起细胞发生G2/M期阻滞.结论在放射免疫导向治疗中,辐射诱导的细胞凋亡与细胞周期发生阻滞,特别是与G2/M期阻滞有着密切的关系,细胞周期阻滞与放射敏感性可能存在正相关.

Abstract:Objective To observe the cell cycle changes of HL60/ADM cells induced apoptosis by ~(131)I-GMCSF.Methods An in vitro radioimmunotherapy (RIT ) model was employed. Data were collected from flow cytometric analysis.Results After exposure to ~(131)I-GMCSF, HL-60/ADM cells were induced to apoptosis and the apoptosis rate increased in the time-dependent manner. The cell cycle showed S arrest and G_2/M arrest after 24h and mostly G_2/M arrest after 48h.Conclusion Cell apoptosis induced by irradiation has close relation to cell cycle arrest especially G_2/M arrest in targeted radioimmunotherapy. Cell cycle arrests may have positive correlation to radiation sensibitity.
