| 摘 要： | 目的]观察痹褀胶囊对实验性骨关节炎(OA)大鼠软骨、关节液及血清基质金属蛋白酶-3(MMP-3)、基质金属蛋白抑制酶-1(TIMP-1)的影响,探讨痹褀胶囊保护关节软骨的作用机制。方法]选Wistar大鼠,木瓜蛋白酶膝关节注射造模,痹祺胶囊灌胃治疗8周,酶联免疫吸附(ELISA)法检测软骨、关节液及血清MMP-3、TIMP-1的活性。结果]痹祺胶囊能提高模鼠软骨TIMP-1的活性(P<0.01),降低关节液MMP-3的活性,使软骨、关节液MMP-3/TIMP-1的比值均降低(P<0.01);对血清MMP-3、TIMP-1及MMP-3/TIMP-1比值无明显影响(P>0.05)。结论]痹褀胶囊保护关节软骨的作用可能是通过提高软骨TIMP-1的活性,降低关节液MMP-3的活性,使软骨、关节液MMP-3/TIMP-1的比值减小,从而抑制MMP-3对关节软骨基质蛋白多糖的裂解活性而起保护作用的。
