摘    要:目的 探讨楤木皂苷对人宫颈癌细胞HeLa细胞增殖和迁移的抑制作用,揭示核因子-κB(NF-κB)在楤木皂苷抑癌过程中的分子机制.方法 以体外培养的人宫颈癌细胞HeLa系为研究对象,实验分为对照组、楤木皂苷(200μg/mL)处理组(观察组),48 h后观察两组HeLa细胞增殖能力的改变.运用Western blot检测NF-κB的表达及酵母自噬基因Atg5/Vps30的同源基因(Beclin 1)、自噬微管相关蛋白轻链3B(LC3B)的改变.结果 楤木皂苷能够显著抑制HeLa细胞的增殖;楤木皂苷能够抑制NF-κB信号通路及促进自噬相关蛋白Beclin 1、Atg 5、LC3B的表达.结论 楤木皂苷能够抑制NF-κB信号通路,进而诱导自噬的发生影响宫颈癌细胞HeLa的增殖和迁移.

Abstract:Objective To explore the inhibition role of araloside on cervical cancer HeLa cell proliferation and migration to reveal the molecular mechanism of NF-κB in HeLa cell in the process of tumor suppression.Methods The in vitro cultured cervical cancer HeLa cell line served as the research object.The experiment was divided into the control group and araloside(200 μg/mL) treatment group(observation group).The change of proliferation and migration ability after 48 h were observed in the two groups.Western blot was used to detect the expression of NF-κB molecule and change of autophagy involved protein Beclin 1 and LC3B.Results Araloside could significantly inhibit the proliferation and migration of HeLa cells and the NF-κB signaling pathway,and promoted the expression of autophagy related molecule Beclin 1,Atg 5 and LC3B.Conclusion Araloside could inhibit the NF-κB signaling pathways,thus induces autophagy occurrence and influences the proliferation and migration of cervical cancer HeLa cells.
