摘    要:目的:摸索改进鸡胚绒毛尿囊膜(chick chorioallantoic membrane,CAM)技术,为抗血管形成药物筛选提供良好的体内血管实验模型.方法:对传统的蛋壳上开窗方式的CAM技术进行改进,将孵育3 d的鸡胚脱离蛋壳移入平皿中,继续孵育3 d后将待测药物植入到CAM和卵黄囊膜(yolk sac membrane,YSM)上,24 h后观察药物对CAM和YSM血管的作用.结果:平皿中孵育鸡胚至第6天,鸡胚存活率为46.7%,生长状况良好,CAM直径达到1.2 cn.检测的6种药物出现两种不同的心血管效应,即抑制和吸引趋化血管的效应.结论:平皿中孵育CAM技术在研究药物、生物因子、肿瘤等的血管效应方面有良好的应用价值.

Abstract:Objective: To improve the technique of chick chorioallantoic membrane (CAM). and establish a good vascular model in vivo for screening antiangiogenesis drugs. Methods: In improving the traditional CAM technique, 3 days'chick embryo was removed out and placed onto plate for further development. After incubating on the plate for 3 days, drugs to be screened were grafted onto CAM and YSM (yolk sac membrane, YSM). The effect, of drugs on CAM and YSM were examined after 24 hours. Results: after 6 days incubation, the survival ratio of chick embryo was 46.7% and growth condition is good. The diameter of CAM reached 1.2cm. The evident responses of antiangiogenesis and chemotaxis were found in the groups of 6 drugs. Conclusion: The technique of incubating CAM in plate have wide application value in researching the vascular effects of drugs, bio factors, tumors, and so on.
