| 摘 要： | 目的探讨中药脑力智宝对PC12细胞凋亡的抑制作用及机制。方法以硝普钠(sodium nitro- prusside,SNP)诱导的PC12细胞凋亡为模型,采用血清药理学方法,用荧光显微镜观察细胞形态学的改变,流式细胞仪检测DNA含量及凋亡细胞百分率,免疫组织化学检测Bcl-2蛋白的表达,以DPH为荧光探针检测细胞膜的流动性。结果脑力智宝提取物及含药血清能减轻SNP引起的细胞核形态学的改变,减少凋亡细胞数,使SNP诱导的PC12细胞凋亡百分率明显下降,凋亡峰显著降低,并能提高Bcl-2蛋白的表达,增加细胞存活率,增加膜流动性。结论脑力智宝对SNP诱导的PC12细胞凋亡具有抑制作用。并可能是通过促进Bcl-2蛋白表达,增加细胞膜流动性等途径而抑制细胞凋亡的。

| Abstract: | Objective To study the inhibitory effect and mechanism of Naoli Zhibao(NLZB)on PC12 cells apoptosis.Methods PC 12 cells apoptosis was induced by sodium nitroprusside(SNP),the DNA content and percentage of apoptosis were measured by flow cytometry;Morphological changes were observed with fluorescence microscope;Bcl-2 protein was detected by immunohistochemical method;Cells viability and membrane fluidity were measured with MTT assay and fluorescence spectrophotometer respectively.Results NLZB and drug-contai- ning serum could relieve the morphological change of nuclei,and prevent the neuronal apoptotic peak induced by SNP.The number of Bcl-2 immunoreactive positive cells was significantly increased by NLZB in cultured PC12 cells.The membrane fluidity of PC12 cells treated by NLZB increased significantly.Conclusion NLZB could in- hibit PC12 ceils apoptosis induced by SNP.The mechanism of NLZB's anti-apoptosis effect may be through promo- ting Bcl-2 protein expression and increasing membrane fluidity. |
