氟西汀对抑郁症大鼠杏仁核磷酸化微管相关蛋白-2表达的影响

摘    要:目的:探讨氟西汀对抑郁症大鼠杏仁核磷酸化微管相关蛋白-2(pMAP-2)表达的影响,为研究氟西汀药效机制提供实验依据。方法:将雄性Wistar大鼠随机分为对照组、模型组和氟西汀组,采用慢性不可预见性温和应激方法建立抑郁大鼠模型,采用糖水偏好实验、悬尾实验、Morris水迷宫实验进行行为学检测,免疫印迹方法检测pMAP-2蛋白表达,RT-PCR法检测pMAP-2 mRNA表达。结果:模型组和氟西汀组大鼠体质量增长率、糖水偏好百分比低于对照组(P<0.05),悬尾不动时间和逃避潜伏期高于对照组(P<0.05);氟西汀组大鼠体质量增长率、糖水偏好百分比高于模型组(P<0.05),悬尾不动时间和逃避潜伏期低于模型组(P<0.05)。模型组和氟西汀组大鼠杏仁核pMAP-2蛋白和mRNA表达高于对照组(P<0.05),氟西汀组低于模型组(P<0.05)。结论:氟西汀能逆转应激抑郁大鼠杏仁核组织中pMAP-2的增高。

Abstract:Objective To observe the effect of fluoxetine on phosphorylated microtubule-associated protein-2(pMAP-2) expression in amygdala of depression model rats,and to provide the experimental basis for the study of efficacy mechanism of fluoxetine.Methods Male Wistar rats were randomly divided into control group,model group and fluoxetine group.The depression animal model was produced by giving the rats chronic unpredicted mild stress.The depressional behavior was examined using sucrose preference test,tail-suspension test and Morris water maze.The expressions of pMAP-2 protein and mRNA were detected using Western blotting and RT-PCR,respectively.Results The changes in body weight(%) and percentage of sucrose preference of rats in model group and fluoxetine group were significantly lower,while the tail-suspension immobility and the excape latency time were higher than those in control group(P<0.05).These indexes were reversed by fluoxetine.The expressions of pMAP-2 protein and mRNA in model group and fluoxetine group were higher than those in control group(P<0.05),while the indexes were decreased after treatment with fluoxetine(P<0.05).Conclusion Fluoxetine can reverse the expression increasing of pMAP-2 in amygdala neurons of depression model rats.
