摘    要:目的制备血管内皮生长因子反义寡核苷酸(VEGF-ASODN)的PLGA纳米粒,并探讨其制备工艺。方法采用复乳化溶剂挥发法制备,并用正交设计法对纳米粒的处方和制备工艺进行优化。结果纳米粒形态圆整,大小均匀,平均粒径150nm,包封率可达72%,含药量0.84%,体外释放缓慢,达到21天。结论VEGF-ASODN的PLGA纳米粒制备工艺简便、重现性好。

Abstract:Objective To study vascular endothelial growth factor (VEGF) antisense oligodeoxynucleotide (VEGF-ASODN) loaded polylactic-co-glycolic acid (PLGA) nanoparticles and its formulation. Methods PLGA nanoparticles were prepared with w/o/w(water/oil/water) double emulsion-solvent evaporation method by orthogonal design. Results The shape of VEGF-ASODN loaded PLGA nanoparticles is roundness, size is well-distributed, the mean diameter is 150 nm, the encapsulation ratio reaches to 72%, the VEGF-ASODN loading in the nanoparticles is 0.84%, and the release is slow for at least 21 days. Conclusion The preparing method of VEGF-ASODN loaded PLGA nanoparticles is simple, and repeatability is fine.
