血府逐瘀汤对实验性脑出血大鼠活化Caspase-3表达的影响

摘    要:目的观察血府逐瘀汤对脑出血模型大鼠颅内血肿周围神经元活化Caspase-3的干预作用。方法用Ⅶ胶原酶注射诱导大鼠脑出血模型,免疫组化检测血肿周围活化Caspase-3的表达。结果Ⅶ胶原酶造模后模型组血肿周围于6h出现Caspase-3阳性表达细胞,1d阳性细胞计数达高峰,以后逐渐下降,与假手术组差异明显(P<0.01)。血府逐瘀汤治疗组组与模型组比较:6h计数无明显差异,1d,3d,5d三个时间点,给药组阳性细胞数量显著减少,细胞着色变浅。(P<0.01)。结论血府逐瘀汤下调脑出血大鼠血肿周围活化Caspase-3的表达,抑制神经细胞凋亡,可能是其疗效机理之一。

Abstract:Objective To explore the neuronal protective effect and its mechanism of a traditional Chinese medicine complex Xuefuzhuyu decoction(XFZYD) on the expression of activated Caspase-3 of experimental intracerebral hemorrhagic rat. Methods Collagenase-induced ICH rats were used as animal models. At the 6h,1d,3d, and 5d time points, Caspase-3 was measured by cell count combined with immunohistochemical method. Results The Caspase-3 positive cells appeared around the hematoma of the model animals at the 6th hour after collagenase treatment and reached at the 1st day, then declined gradually. The decline was slight at the 3rd day, and became evident at the 5th day as compared with the control group, the difference showed statistical significance (P<0.01). The number of positive cells of the XFZYD group was less than that of the model group at the 1d, 3d, and 5d time points (P<0.01), but not at the 6th hour. The activated Caspase-3 of treated group was less than those of the model control group (P<0.01). Conclusions The neuronal protective mechanism of Xuefuzhuyu decoction following ICH may be related with its reduction effect of activated Caspase-3, which contributes to inhibiting cellular apoptosis and protecting the function of neurocyte.
