摘    要:目的探讨凋亡相关基因caspase-3和bcl-2在大肠腺瘤及大肠腺癌组织中的表达及其意义。方法采用高通量的组织芯片技术及免疫组织化学S-P法检测115例大肠腺癌组织、19例大肠腺瘤组织和12例正常大肠组织中caspase-3和bcl-2的表达。结果在正常大肠黏膜、腺瘤和大肠腺癌组织中caspase-3的阳性表达率分别为83.33%、47.37%、41.74%,bcl-2的阳性表达率分别为25%、84.21%、58.26%;正常黏膜组织caspase-3的表达高于大肠腺瘤和大肠腺癌,而bcl-2表达低于后两者,差异均有统计学意义(均P〈0.05)。caspase-3和bcl-2的阳性表达率与大肠腺癌患者的组织分化程度相关(P〈0.05)。结论组织芯片技术是大规模平行检测多基因蛋白表达的一种有效方法。caspase-3和bcl-2可能在大肠腺癌的发生、发展中起了重要作用。

Abstract:Objective To investigate the expression and significance of apoptosis-related gene caspase-3 and bcl-2 in colorectal adenoma and adenocarcinoma. Methods Using tissue microarrays and immunohistochemical staining,the expression of caspase-3 and bcl-2 was detected in 12 cases of normal mucosa, 19 cases of colorectal adenoma and 115 cases of colorectal adenocarcinoma. Results The positive expression of caspase-3 in normal mucosa was 83.33% higher than 47.37% in adenoma and 41.74% in cancer specimens (P〈0.05).The positive expression of bcl-2 in normal mucosa was 25%, lower than 84.21% in adenoma and 58.26% in cancer specimens (P〈0.05).The expression of caspase-3 and bcl-2 was significantly correlated with degree of differentiation (P〈0.05). Conclusion Tissue microarray technique is an effective method to detect multiple gene protein expression. Caspase-3 and bcl-2 may play an important role in genesis development and metastasis of colorectal adenocarcinoma.
