Skp2和P27kip1蛋白在胰腺癌中的表达及与临床病理特征的关系

摘    要:目的:探讨S期激酶相关蛋白2(S-phase kinase-associated protein 2,Skp2)与P27kip1在胰腺癌中的表达及与临床病理特征的关系。方法:采用免疫组化方法(SP法)检测31例胰腺癌组织和14例癌旁组织中Skp2和P27kip1的表达。结果:31例胰腺癌组织中Skp2阳性表达率为32.26%(10/31),显著高于癌旁组织0%(0/14)(P<0.05);P27kip1的阳性表达率为45.16%(14/31),明显低于癌旁组织78.57%(11/14)(P<0.05)。统计分析还显示:随着癌组织分化程度降低、TNM分期升高、淋巴结的转移Skp2阳性表达率相应升高(P<0.05),而P27kip1的阳性表达率降低(P<0.05)。两者的表达与性别、年龄及肿瘤大小均无关(P>0.05),且Skp2和P27kip1在胰腺癌的表达存在明显的负相关(rs=-0.59,P<0.01)。结论:Skp2和P27kip1在胰腺癌发生、发展中有重要作用。

Abstract:Objective: To investigate the relationship of S-phase kinase-associated protein 2(Skp2) and P_(27kip1)expressions with clinicopathological features of pancreatic cancer.Methods: S-P immuno- histochemical technique was applied to detect the expressions of Skp2 and P27~(kip1) proteins in tumor tissues of 31 patients and adjacent tissues of 14 patients with pancreatic cancer.Results: Skp2 protein positive-expression rate in tumor tissues of pancreatic cancer was 32.56%(10/31),which was higher than that in adjacent tissues 0%(0/14)(P<0.05);P_(27kip1)protein positive- expression rate in tumor tissues of pancreatic cancer was 45.16%(14/31),which was lower than that in adjacent tissues,which was 78.57%(11/14)(P<0.05).Skp2 protein positive-expression rate correlated remarkably with poorly differentiated tumor cells,high TNM stages or lymph node metastasis(all P<0.05).But P_(27kip1)protein positive-expression rate was lower(P<0.05),and had no relation with other clinicopathological features including sex,age and size of tumor(all P>(0.05).) Expression of Skp2 was negatively correlated with expression of P27~(kip1)(r_(s)=-0.59,P<0.01).(Conclusion:) Skp2 and P27~(kip1) play an important role in the oncogenesis and development of pancreatic cancer.
