pSVPoMcat微基因修饰雪旺氏细胞移植对脊髓损伤c-fos、bcl-2基因表达的影响

摘    要:目的探讨pSVPoMcat微基因修饰雪旺氏细胞(SC)移植对脊髓损伤(SCI)后c-fos,bcl-2基因表达的影响.方法用切割法制备大鼠SCI模型,将实验动物分为pSVPoMcat微基因修饰SC组(A组),SC移植组(B组),损伤组(C组),正常对照组(D组),不致伤.用地高辛(DIG)标记的c-fos、bcl-2探针行原位杂交.结果SCI后A、B、C、D四组脊髓c-fos基因表达顺序为C>B>A=D;bcl-2基因表达顺序为D=A>B>C.结论SCI后C-fos基因表达显著增多,而bcl-2基因表达显著减少,pSVPoMcat微基因修饰SC移植能部分抑制SCI后c-fos基因的表达和促进bcl-2基因的表达.

Abstract:objective: To study the effects of microgene pSVPoMcat implanted to modify Schwann cell (SC) on c - fos and bcl - 2 gene expression after spinal cord injury (SCI) in rats. Method: The animals were divided into four groups: the group of microgene pSVPoMcat implanted to modify SC (group A), SC implanted group (group B), injured group (group C) and uninjured control group (group D) . The levels of expression of c - fos and bcl - 2 genes were determined by situ hybridization after operation 8 hours. Result: The c - fos gene expression increased significantly and bcl - 2 gene expression decreased significantly in group C, as compared to those of group D (p < 0.05) . Although c - fos gene expression increased and bcl - 2 gene espression decreased in group A, the changes were not obvious as in group C (p < 0.01) . Conclusion: The c - fos gene expression could be induced and bcl - 2 gene expression be inhibited following SCI. The microgene pSVPoMcat implanted to SC might play some role in inhibition of c - fos gene expression and promotion of bcl - 2 gene expresion following SCI.
