MicroRNA-221通过抑制CDKN1C/p57表达促进结肠癌细胞增殖

摘    要:目的探讨microRNA-221(MIR221)对结肠癌细胞中CDKN1C/p57表达调控及细胞增殖的影响。方法常规培养人结肠癌Caco2细胞系,预先给予或不给予CDKN1C/p57干扰性小RNA(anti-p57-siRNA)处理,再以脂质体分别转染MIR221前体(pre-MIR221)或MIR221特异性抑制剂(anti-MIR221)寡核苷酸,应用实时荧光定量PCR(Real-time RT-PCR)检测Caco2细胞中MIR221表达状况,运用半定量RT-PCR及Western-blot分析CDKN1C/p57 mRNA及蛋白表达状况,并通过噻唑蓝(MTT)比色法观察细胞的增殖状态;构建pGL3-p57荧光素酶报告载体并与pre-MIR221或anti-MIR221共转染Caco2细胞,检测转染细胞中荧光素酶活性变化。结果 Caco2细胞转染pre-MIR221后,CDKN1C/p57蛋白表达水平下降,细胞增殖显著(P<0.01);而anti-MIR221可上调CDKN1C/p57蛋白表达继而抑制细胞增殖,且此种抑制作用可被anti-p57-siRNA逆转,说明此种抑制效应确由CDKN1C/p57所介导;在转染重组有CDKN1C/p57基因3’-UTR片段的荧光素酶报告载体的Caco2细胞中,共转染pre-MIR221后荧光素酶活性下降,而共转染anti-MIR221后荧光素酶活性增高,差异均具有统计学意义(P<0.01)。结论 MIR221可通过与CDKN1C/p57 mRNA 3’-UTR区域结合使结肠癌细胞中CDKN1C/p57蛋白表达下降而促进肿瘤增殖;anti-MIR221则可通过上调CDKN1C/p57蛋白表达继而抑制细胞增殖而发挥抗瘤效应。
