摘    要:目的探讨8-羟基二氢小檗碱对NIT-1细胞胰岛素分泌的影响及其机制。方法采用不同浓度8-羟基二氢小檗碱干预NIT-1细胞后,以放射免疫法、液体闪烁计数法、酶法,分别检测其胰岛素水平、葡萄糖利用、葡萄糖激酶(GK)活性。结果在低浓度葡萄糖(5.5mmol/L)刺激下,与空白对照组相比,加入高浓度8-羟基二氢小檗碱(1.0μmol/L)的NIT-1细胞胰岛素分泌增加、葡萄糖利用活跃,且GK酶活性增强(P<0.05);在高浓度葡萄糖(16.5mmol/L)刺激下,与空白对照组相比,加入不同浓度8-羟基二氢小檗碱(0.5、1.0μmol/L)的NIT-1细胞胰岛素分泌增加、葡萄糖利用活跃,且GK酶活性增强(P<0.01或P<0.05)。结论 8-羟基二氢小檗碱能促进葡萄糖刺激下NIT-1细胞的胰岛素分泌,其机制可能与增加NIT-1细胞葡萄糖利用、增强GK酶活性有关。
