5-氮杂-2'-脱氧胞苷和曲古霉素A联合应用对胶质瘤细胞增殖和侵袭的作用

摘    要:目的胶质瘤的化疗一直都是研究热点,近来有研究表明甲基化与胶质瘤的发生和发展关系密切,本研究的目的是探讨去甲基化试剂5-氮杂-2'-脱氧胞苷(5-Aza-2'-deoxycytidine)和曲古霉素A(TSA)联合应用对胶质瘤细胞系侵袭和转移的调节。方法分别将浓度为50μM的5-氮杂-2'-脱氧胞苷和300ng/ml的曲古霉素A加入胶质瘤细胞系U251细胞中,并将两者联合应用加入U251细胞系中,应用MTT法检测不同处理组细胞的增殖能力,Transwell细胞侵袭实验检测不同处理组和对照组中细胞的侵袭能力的变化。结果 5-氮杂-2'-脱氧胞苷和曲古霉素A单独处理后,U251细胞的增殖能力明显减弱(<0.05),两者联合应用时对U251细胞的增殖抑制更明显(<0.01)。Transwell细胞侵袭实验显示5-氮杂-2'-脱氧胞苷和曲古霉素A单独处理后,U251细胞的侵袭细胞数明显减少(<0.05),而两者联合应用时对U251细胞的侵袭能力抑制更为明显(P<0.01)。结论 5-氮杂-2'-脱氧胞苷和曲古霉素A都可以抑制胶质瘤细胞系的增殖和侵袭,两者具有协同作用。

Abstract:Objective The chemical therapy is always a hot spot of glioma study.Many studies have shown that DNA methylation closely correlated with the occurrence and development of glioma,the purpose of this study is to investigate the effect of combined 5-Aza-2'-deoxycytidine and TSA treatment on the proliferation and invasion of glioma cell line U251.Methods 5-Aza-2'-deoxycytidine(50μM) and Trichomycin A(TSA,300ng/ml) was added into U251 cells,respectively.Combined use of 5-Aza-2'-deoxycytidine and TSA was added into U251 cells.MTT method was applied to determine the proliferation ability and transwell cell invasive assays was used to determine the invasion ability in U251 cells.Results 5-Aza-2'-deoxycytidine or TSA treatment could down-regulate the proliferation of U251 cells(<0.05),the combined use of 5-Aza-2'-deoxycytidine and TSA could down-regulate the proliferation of U251 cell more obviously than 5-Aza-2'-deoxycytidine or TSA single treatment.Transwell cell invasive assays showed that 5-Aza-2'-deoxycytidine or TSA treatment could downregulate the invasion of U251 cells(<0.05),the combined use of 5-Aza-2'-deoxycytidine and TSA could down-regulate the invasion of U251 cell more obviously than 5-Aza-2'-deoxycytidine or TSA single treatment.Conclusion 5-Aza-2'-deoxycytidine or TSA treatment could inhibit the proliferation and invasion of glioma cell line U251,they have cooperative effect on the proliferation and invasion of glioma cell line U251.
