水蛭素抑制体外模拟增生性瘢痕模型的实验研究

摘    要:目的:研究水蛭素对体外人增生性瘢痕模型的收缩能力,成纤维细胞的增殖、胶原分泌等功能的影响。方法:体外培养人增生性瘢痕成纤维细胞(HSFB),建立含增生性瘢痕成纤维细胞的胶原网架系统(FPCL),用1、10、50U/ml的水蛭素溶液干预该模型,测定凝胶块收缩指数,利用MTT染色计数活细胞,逆转录多聚酶链式反应(RT-PCR)法检测细胞Ⅰ、Ⅲ型前胶原mRNA表达。结果:浓度为10U/ml和50U/ml水蛭素溶液能够抑制HSFB的活力,使FPCL收缩指数降低,抑制FPCL中HSFBⅠ、Ⅲ型前胶原mRNA的表达(P<0.05)。结论:水蛭素溶液可抑制体外人增生性瘢痕三维模型的收缩功能和成纤维细胞增殖、分泌胶原功能。
