靛氰绿在四氯化碳染毒大鼠肝胆内转运的研究

摘    要:目的 研究靛氰绿 (ICG)在四氯化碳 (CCl4 )致急性肝损伤大鼠的 (CCl4 大鼠 )肝胆内转运过程。方法 复制CCl4 大鼠模型 ,观察下列变化 :(1)各种病理学的变化 ;(2 )肝血流量和靛氰绿肝清除率的改变 ;(3 )靛氰绿的药代动力学参数。结果 靛氰绿在CCl4 染毒大鼠体内的清除减慢 ,总清除率下降至对照组的约 70 % (P <0 0 5 ) ;肝清除率和肝血流量也明显下降 (P <0 0 5 )。结论 靛氰绿在CCl4 染毒大鼠的肝清除率下降 ,药代动力学分析 ,结果提示至少与靛氰绿自血液进入肝细胞速度减慢 ,以及肝血流量的减少有关。

Abstract:Objective To study the hepatobiliary transportation of indocyanine green(ICG)in normal rats and rats-treated by carbon tetrachloride(CCl\-4).Methods CCl\-4-treated rats(CCl\-4-rats)were experimentally prepared by a single oral administration of CCl\-4.The following studies were carried out in normal and CCl\-4-rats:(1)Observation of the pathophysiological changes;(2)determination of hepatic plasma flow and ICG hepatic clearance by a continuous ICG infusion method;and (3)analysis of pharmacokinetic parameters and hepatobiliary transportation of ICG after intravenous administration.Results In CCl\-4-rats,the elimination of ICG was prolonged and the total plasma clearance was about 70% of the control value( P <0.05).The hepatic clearance of ICG,hepatic plasma flow was also decreased in CCl\-4-rats( P <0\^05). Conclusion The total plasma clearance of ICG was significantly decreased in CCl\-4-rats.The pharmacokinetic analysis of these data indicates that the decreased total plasma clearance of ICG in CCl\-4-rats is owing to at least two factors:(1)defective uptake of ICG by hepatocytes and (2) the decrease in hepatic plasma flow.
