SATB-1基因在前列腺癌中的特异表达及临床意义

摘    要:目的 探讨特异性核基质结合区结合蛋白(SATB-1)在前列腺癌中的表达及临床意义.方法 收集前列腺增生、前列腺癌无骨转移、前列腺癌骨转移各30例患者的前列腺组织标本,采用免疫组织化学S-P法检测SATB-1的表达,并结合临床病理因素进行分析.结果 SATB-1表达阳性率在前列腺增生组织为0(0/30),前列腺癌组织为86.7%(52/60),两组比较差异有统计学意义(P<0.05);前列腺癌Gleason评分2~4分者SATB-1表达阳性率为76.5%(13/17),5~7分者为85.0%(17/20),8~10分者为95.7%(22/23),组间比较差异无统计学意义(P>0.05).采用双评分半定量法对SATB-1表达强度进行评分,经秩和检验SATB-1表达强度在前列腺癌无骨转移组与前列腺癌骨转移组间比较差异有统计学意义(P<0.05).SATB-1表达强度在前列腺癌不同Gleason评分组间比较差异有统计学意义(P<0.05),表达强度随Gleason评分的增加而增强.结论 SATB-1仅在前列腺癌组织中表达,且在Gleason评分高、有骨转移的前列腺癌组织中表达强度明显增加.SATB-1有望作为前列腺癌诊断和判断预后的有效指标.

Abstract:Objective To investigate the expression of special AT rich sequence binding protein (SATB-1) in prostatic carcinoma.Methods Expression of SATB-1 was examined in 90 paraffin-embedded tissue sections including 60 cases of prostatic carcinoma and 30 cases of benign prostate hyperplasia by immunohistochemical staining.The relationship between SATB-1 expression and clinicopathological features was analyzed by Chi-square test.Results The immunohistochemistry revealed that the positive rate of SATB-1 in 60 of prostatic carcinoma was 86.7% (52/60),30 of benign prostate hyperplasia was 0 (0/30).The score of staining intensity of SATB-1 in benign prostate hyperplasia samples,prostatic carcinoma samples without bone metastasis and with bone metas tasis were gradually increasing significantly.The difference among the pathological Gleason score was statistically significant.Conclusion Expression of SATB-1 has close relations with the occurrence and development of prostatic carcinoma.It is hopeful that SATB-1 can be used as one of the indexes to predict early metastasis.
