牙龈卟啉菌核酸探针的制备,鉴定

摘    要:根据Pg的特异的fimA基因设计一对引物,采用PCR技术扩增其中542bp长的DNA片段,用~(32)P标记作为探针与25株细菌DNA进行杂交鉴定.结果显示该探针能识别5株同种菌,而与其它20株异种菌DNA无阳性信号出现,探针的敏感性达到微微克级.提示该探针具有特异、敏感、快速、简便的特性.适用于对Pg的检测.

Abstract:In the present study, the primers were designed on the basis of the specific fimA gene of Pg. The specific DNA fragments of 542-bp amplified by PCR were used as a 32P-labelled probe hybridizing and identifying with DNA of 25 strains of bacteria. The results indicated that the probe could distinguish 5 strains of Pg, while 20 strains of heterologous species failed to show the positive signals of the hybridization. The sensitivity of the probe reached the level of Pg. This demonstrates that the probe is of the characterization of the specificity, sensitivity,rapidity,simplicity. It is helpful to detect Pg.
