摘    要: 目的 建立适合盐酸西替利嗪血浆浓度测定的反相高效液相色谱法(RP-HPLC)。方法 选择离子对反相高效液相色谱法,以乙腈-0.02 mol·L-1磷酸二氢钠-三乙胺(50∶50∶0.15,内含十二烷基磺酸钠0.007 mol·L-1)为流动相,在波长229 nm处检测。结果 在10~400 ng·ml-1的浓度范围内血浆中西替利嗪具有良好的线性关系,平均萃取回收率大于70%,日内日间精密度均小于5%。结论 本法适用于测定血浆中西替利嗪的浓度,并进一步用于研究西替利嗪在人体内药代动力学过程。

Abstract:OBJECTIVE To establish a RP-HPLC method for the determination of cetirizine in plasma. METHODS Cetirizine was determined by a reversed-phase ion-paired high-performance liquid chromatography with the detection wavelength at 229 nm.The mobile phase was ACN:0.02 mol·L -1phophate:TEA 50∶50∶0.15),containing 0.007 mol·L-1SDS. RESULTS The calibration curve was linear within the concentration range of 10~400 ng·ml-1of cetirizine in plasma.The mean extraction recovery was more than 70% and the relative standard deviations for the within-day and the between-days variation were all less than 5%. CONCLUSION This new method could be used to determine cetirizine in plasma and will be suitable to the pharmacokinetic study of cetirizine.
