摘    要:目的改进大鼠神经干细胞的分离和培养方法。方法用出生1~3d的新生大鼠,取脑用刀片剁碎和吸管吹打的机械分离法代替酶消化法。用免疫细胞化学法鉴定神经干细胞、神经元和星形胶质细胞。结果用机械分离法从新生大鼠脑组织分离神经干细胞的获得率较高,并培养出较多的神经球,还能分化成神经元和星形胶质细胞。结论该方法简便,能得到较多的神经球,细胞获得率也较高。

Abstract:Objective To modify the isolation and culture method of neural stem cells from rats. Methods Neonatal (1 ~ 3 d after birth ) rats were used . The neural stem cells were isolated with mechanical dispersion method in stead of enzymatic digestion method. The immunocytochemistry was used to identify the neural stem cells, neurons and astrocytes. Results More neurospheres and high cell harvest rate were obtained from neonatal rat brain with mechanical dispersion method. The isolated cells could differentiate to neurons and astrocytes. Conclusion This isolation and culture method is simple, the number of neurospheres is more and the cell havest rate is higher.
