流式细胞血小板交叉配型试验方法的建立

摘    要:目的建立检测灵敏度更高的流式细胞血小板交叉配型(Flow-XM)的试验方法。方法对104名临床血小板输注>3次的患者作Flow-XM试验,同时采用经典补体依赖微量淋巴细胞毒交叉配型(NIH-CDC)方法作对照,比较2种方法的试验结果。结果血小板阳性检出率,Flow-XM试验为60.58%(63/104),NIH-CDC试验为26.92%(28/104)(P<0.01)。结论 Flow-XM试验具有较高的灵敏度,能够对原始结果全程质控,可使血小板交叉配型技术实现了标准化。

Establishment of the experiment method for flow cytometry of platelet cross match

Abstract:Objective To set up a more sensitive detection method of platelet cross match using flow-cytomerty.Methods Blood samples from 104 patients who had received platelet transfusion more than three times underwent Flow-XM and NIH-CDC detection.The results between two different techniques were compared.Results In 104 patients,63 were positive in Flow-XM(60.58%),28 were positive in NIH-CDC(26.92%),there was significant difference between two methods(P<0.01) Conclusion Flow-XM is more sensitive than NIH-CDC,additionally,the computer printouts represent a permanent record of the cross match for retrospective review.Flow-XM may become the standard cross match method as a possible alternative to be recommended.
