摘    要:①目的建立糖尿痛心肌损伤动物模型,探讨糖尿病心肌损伤的相关因素。②方法用高脂饲料喂养和腹腔注射链脲佐菌素诱发大鼠2型糖尿病胰岛素抵抗模型,检测血糖和糖耐量、血清TC、TG、血浆胰岛素、心肌组织TNF—a、AngⅡ、AGEs的含量和心脏重量指数,光镜下观察心肌形态结构变化。③结果糖尿病模型组空腹血糖和糖耐量各点血糖值、血清TC、TG、血浆胰岛素、心肌组织TNF—a、AngⅡ、AGEs的含量和心脏重量指数均较正常对照组明显增高(P〈0.01),光镜下见糖尿病大鼠心肌细胞明显肿胀肥大、变性,心肌纤维结构紊乱。④结论高脂饲料喂养联合腹腔注射STZ可建立2型糖尿病心肌损伤的大鼠模型,可能与心肌局部AngⅡ、TNF—a和AGEs过度表达有关。

Experimental study on myocardial injury effect of type 2 diabetes insulin resistance

Abstract:Objective To explore the related factors of diabetic myocardial injury by establishing animal model of type 2 di abetes insulin resistance. Methods The model was made by fat - enriched diet and intraperitoneal injection of streptozotocin. (STZ). The level of fasting blood glucose (FBG) was detected and oral glucose tolerance test (OGTY) was performed. The con tent of serum TC, TG, plasma insulin, myocardial tissue TNF - a, Ang Ⅱ AGEs were determined. The heart weight index was evaluated and changes in myocardial mo'rphology were observed. Results Compared with control group, the level of FBG and blood glucose of each point in OGTF, and the content of serum TC, TG, plasma insulin, myocardial tissue TNF - ct, Ang Ⅱ , AGEs in creased remarkably in model group ( P 〈 0.01 ). The cardiomyocytes of diabetes rats looked obviously swelled, hypertrophy and degenerated, and the structure of myocardial fiber was disordered under light microscope. Conclusion The myocardial injury mod- el of type 2 diabetes insulin resistance in rats , which might be related to the over - expressior/of local Ang Ⅱ ,TNF - a and AGEs in myocardial tissue, could be established with fat - enriched diet and intraperitoneal injection of STZ.
