RIN1蛋白在卵巢癌细胞和组织中的表达及其临床病理意义

摘    要:目的 探讨ras及rab作用因子1(RIN1)蛋白在卵巢癌细胞的表达水平及定位,并探讨其在卵巢肿瘤组织中的表达水平及临床意义。方法 采用免疫印迹(Western blotting)法检测RIN1蛋白在人正常卵巢上皮细胞及卵巢癌细胞株OVCAR3、SKOV3和HO8910中的表达水平及定位;检测其在17例卵巢正常组织和91例卵巢肿瘤组织中的表达并分析其与卵巢癌分期和组织分化程度的关系。结果RIN1蛋白在卵巢癌细胞 OVCAR3、SKOV3和HO8910中的相对表达量(0.3641±0.0614、0.6454±0.0835和0.7925±0.1491)高于人正常卵巢上皮细胞(0.0679±0.0269),差异有统计学意义(P<0.05),且主要分布于卵巢癌细胞胞浆中。RIN1蛋白在37例良性卵巢肿瘤、10例交界性卵巢肿瘤及44例恶性卵巢肿瘤组织中相对表达量分别为0.3456±0.1870、0.5587±0.2138和0.7236±0.2232,均高于其在17例卵巢正常组织中的表达水平(0.1284±0.0982),差异有统计学意义(P<0.01),且RIN1蛋白的表达随卵巢病变程度的加重而升高;RIN1蛋白表达随卵巢癌手术、病理分期及组织分化程度的演进而增加。结论 RIN1蛋白在卵巢癌细胞中高表达且定位于胞浆中;RIN1蛋白高表达与卵巢癌发生发展密切相关,其可作为卵巢癌早期诊断的一个新的肿瘤标志物,并有望成为卵巢癌治疗的新靶点。

Abstract:Objective To investigate the expression and location of ras and rab interactor 1 (RIN1) in ovarian cancer cell lines and analyze the expression and clinicopathological significance of RIN1 protein in ovarian cancer tissues. Methods Expression and location of RIN1 protein in normal human ovarian epithelial cell line and three ovarian cancer cell lines ( OVCAR3, SKOV3 and HO8910) were detected by Western blotting assay as well as the expression of RIN1 protein in 17 cases of normal ovarian tissues and 91 cases of ovarian tumor tissues. Results RIN1 protein in ovarian cancer cell lines OVCAR3, SKOV3 and HO8910 (0. 3641±0. 0614, 0. 6454±0. 0835 and 0. 7925±0. 1491 ) were higher than that in normal human epithelial cell(0. 0679±0. 0269) with statis- tical significance( P 〈 0. 05 ) , and RIN1 protein localized chiefly in the cytoplasm of ovarian cancer cell lines. The expressions of RIN1 protein in 37 benign ovarian tumors, 10 borderline ovarian tumors and 44 malignant tumors were 0. 3456±0. 1870, 0. 5587±0. 2138 and 0. 7236±0. 2232, much higher than that in 17 normal ovarian tumor tissues (0. 1284±0. 0982) with significance (P 〈 0. 01 ). The expression of RIN1 increased with the severity of ovarian tumors. The expression of RIN1 protein was significantly increased with the progression of surgical pathological stage and tumor differentiation. Conclusion RIN1 protein was overexpressed in ovarian cancer cells and localized chiefly in the cytoplasm of cancer cell. Overexpression of RIN1 was associated with tumor progression in ovarian cancer, and RIN1 may act as a new tumor marker for diagnosis and a new therapeutic target in patients with ovarian cancer.
