饱和氢气盐溶液对高龄大鼠肾脏衰老的影响

摘    要:目的:观察饱和氢气盐溶液对高龄大鼠肾脏衰老的影响。方法:取10只健康雄性成年SD大鼠,作为成年组;另取20只健康雄性高龄SD大鼠,随机均分成高龄组和饱和氢气盐溶液组(H2组)。成年组及高龄组以5 mL.kg-1.d-1生理盐水灌胃,H2组以5 mL.kg-1.d-1饱和氢气盐溶液灌胃,其他条件等同。8周后检测各组大鼠肾脏组织中超氧化物歧化酶(superoxide dismutase,SOD)、丙二醛及8-羟基脱氧鸟苷(8-hydroxy-2'-deoxyguanosine,8-OHdG)的含量,采用RT-PCR观察肾脏组织中p53,p16,沉默信息调节因子1(silence information regulator 1,SIRT1)的表达水平。结果:与成年组相比,高龄组SOD活力明显降低,丙二醛和8-OHdG的含量明显升高(P〈0.01或P〈0.05);与高龄组相比,H2组SOD活力明显升高,丙二醛和8-OHdG明显降低(P〈0.01或P〈0.05)。与成年组相比,高龄组p53和p16 mRNA表达量均明显升高,SIRT1明显降低(P〈0.01或P〈0.05);与高龄组相比,H2组p53和p16 mRNA表达量明显降低,SIRT1明显升高(P均〈0.01)。结论:饱和氢气盐溶液能够发挥延缓肾脏衰老的作用,可能与抑制p53,p16的表达而促进SIRT1的表达有关。

Abstract:Objective: To observe the impact of hydrogen-rich saline on the senescence of kidney in aging rats.Methods: Ten healthy adult male Sprague-Dawley rats aged 3-4 months were selected in the study as the adult group;meanwhile,twenty healthy male SD rats aged 18-20 months were randomized into aging group and hydrogen-rich saline group.Adult group and aging group rats received intragastric administration of saline(5 mL.kg-1.d-1),while hydrogen-rich saline group rats received hydrogen-rich saline(5 mL.kg-1.d-1).The other conditions were the same in the three groups.After eight weeks,the superoxide dismutase(SOD),malondialdehyde(MDA),8-hydroxy-2'-deoxyguanosine(8-OHdG) contents in kidney tissues from all the rats were determined;and the mRNA levels of p53,p16,SIRT1 were measured by RTPCR.Results: Aging group had greatly lower SOD contents,higher MDA and 8-OHdG concentrations than adult group;while hydrogen-rich saline group had significantly higher SOD contents,lower MDA and 8OHdG concentrations than aging group(P 0.01 or P 0.05);Compared with adult group,aging group had significantly higher p53 and p16 mRNA levels with lower SIRT1 mRNA levels(P 0.01 or P 0.05);on the contrary,hydrogen-rich saline group had markedly lower p53 and p16 mRNA levels with higher SIRT1 mRNA levels,compared with aging group(both P 0.01).Conclusion: Hydrogen-rich saline could play a role in delaying kidney senescence,which was probably related to the inhibition of p53 and p16 expression and the improvement of SIRT1 expression.
