5-羟色胺转运体基因多态与强迫症的关系

目的 探索上海地区汉族人口中5-羟色胺转运体基因（SLC6A4）多态与有／无强迫人格的强迫症的关系。方法 采用聚合酶链反应扩增片段长度多态（PCR—AmFLP）技术，测定120例强迫症（OCD）患者和130例健康对照者的基因型；根据SCID—Ⅱ对强迫症患者与人格障碍共病情况的评定结果进行亚组划分。结果 强迫症患者5-HTTLPR、5HTT—VNTR基因型分布与健康对照者之间存在显著差异（P〈0．05）；不伴有强迫型人格者5-HTTLPR、5HTT-VNTR基因型和等位基因分布与对照者之间存在非常显著性差异（P〈0．01）。结论 在上海汉族人群中SLC6A4基因可能与无强迫型人格障碍的强迫症存在关联；第二内含子的等位基因10和12／10基因型、启动子的L／L基因型可能是此型强迫症的风险因子；有／无强迫人格的强迫症患者可能有着不同的发病机制。

Relation Between Polymorphism of Serotonin Transporter Gene and Obsessive-Compulsive Disorder

Objective To explore the relation between polymorphism of serotonin transporter gene (SLC6A4) and obsessive-compulsive disorder(OCD) with or without obsessive-compulsive personality disorder(OCPD)in the Han nationality of Shanghai. Methods One hundred and twenty OCD patients and 130 healthy subjects were included in this study. All subjects were genotyped directly via polymerase chain reaction amplification fragment length polymorphism techniques. SCID-II was used to evaluate the comorbidity of OCD and personality disorders. ResultsThere was significant difference of genotype of intron 2 (5HTT-VNTR) and promoter(5-HTTLPR)of SLC6A4 gene between patients and controls(P<0.05). There was very significant difference of genotype and allele frequence of 5HTT-VNTR and 5-HTTLPR between non-OCPD patients and controls(P<0.01). Conclusion The polymorphism of SLC6A4 gene may be associated with OCD subjects without OCPD in the Han nationality. With allele 10, 12/10 genotype of 5HTT-VNTR or L/L genotype of 5-HTTLPR may be risk factors for this type of OCD. Patients with or without OCPD may have different etiology.
