PCR-反相杂交法检测沙门氏菌invA、invE基因

摘    要:目的:探讨从血液中早期检出伤寒沙门氏菌病原体的方法。方法:选择沙门氏菌属中的invA invE,采用PCR扩增-反相杂交法及电泳法同时对血培养物中的伤寒沙门氏菌进行对比检测。结果:电泳法6h检出8份,检出率80%。8h、10h可将10份全部检出,检出率为100%。反相杂交法4h有3份阳性,阳性率为30%,6h有9份出现阳性反应,阳性率为90%,8h、10h可将10份全部检出,阳性率为100%。临床标本中电泳法12/16阳性,阳性率75%,PCR-反相杂交法阳性14/16,阳性率87.5%。结论:反相杂交法具有敏感性高、特异性好等优点,对伤寒的早期快速诊断及治疗有重要意义。

Abstract:Objective:[WT5BZ]To explore the methods to detect bacterium typhosum of blood in early stage.[WT5HZ]Methods:[WT5BZ]InvA and invE gene of salmonelleae in blood culture material were examined by PCR-reversed hybridization and electrophoresis method.[WT5HZ]Results:[WT5BZ]Of 8(80%)and 10(100%) samples could be detected positively by agarose electrophoresis method respectively after 6 h and after 8 h and 10 h. However,3(30%) samples were detected positively by reversed hybridization after 4 h and 9(90%) and 10(100%)samples were positive after 6 h and 8 h.The positive findings of clinical specimen were 12/16(75%)and 14/16(87.5%)respectively by agarose electrophoresis and by PCR-reversed hybridization method.[WT5HZ]Conclusion:[WT5BZ]The results show that PCR-reversed hybridization method is obviously superior to agarose electrophoresis for the detecting of invA and invE gene.The method has high sensibility.specificity and reliability and has certain significance for the early diagnosis and treatment of typhoid.
