息痛颗粒对小鼠出凝血时间及血瘀型大鼠血小板聚集的影响

摘    要:目的:观察息痛颗粒对小鼠出凝血时间及对血瘀型大鼠血小板聚集的影响.方法:小鼠50只,随机分为空白对照组、阳性对照组,息痛颗粒大、中、小剂量组,每组10只,分别以生理盐水,丹参滴丸0.1g/(kg·d),息痛颗粒6.26g/(kg·d)、3.13g/(kg·d)、1.25g/(k·d)灌胃给药,1次/d,连续3 d.于末次灌胃后断尾法测出血时间,用毛细玻管法和玻片法测凝血时间.大鼠60只,随机分成6组,每组10只.血瘀模型组、阳性对照组、息痛颗粒大、中、小剂量组分别以生理盐水,丹参滴丸37.5 mg/(k·d),息痛颗粒5g/(k·d)、2.5g/(k·d)、1.25g/(kg·d)灌胃给药,1次/d,连续3 d.以上5组均于末次给药后1 h用肾上腺素加冷刺激形成大鼠急性"血瘀"模型.以正常大鼠灌服生理盐水作空白对照.第2 d各组分别自颈动脉取血,以比浊法测二磷酸腺苷(ADP)、胶原(COL)、花生四烯酸(AA)诱导的血小板最大聚集率.结果:与空白对照组比较,息痛颗粒大、中剂量组小鼠出、凝血时间均延长(P均<0.05);息痛颗粒各组血瘀型大鼠ADP、COL、AA诱导的血小板最大聚集率均降低(P均<0.05).结论:息痛颗粒有抗血小板聚集的功能.

Abstract:Aim:To observe the effects of Xi Tong Ke Li on platelet aggregation in blood stagnation rats,and bleeding time (BT) and coagulation time(CT) in mice.Methods:Fifty mice were randomly allocated into normal control group, positive control group (Dan Shen Di Wan), and Xi Tong Ke Li low-, middle-, and high dose groups, 10 mice in each group. BT and CT were determined after 3 days administration; sixty rats were randomly allocated into model group, positive control group, Xi Tong Ke Li low-, middle-, and high-dose groups and normal control group. After 3 days administration, the first 5 groups were established blood-stagnation model by injecting adrenalin and bathing in ice water.The rate of platelet aggregation induced by ADP,COL,AA was determined.Results: Compared with those of the normal control,the platelet aggregation rates of rats in 3 Xi Tong Ke Li groups in high- and middle-dose Xi Tong Ke Li groups were lower ( P<0.05); BT and CT of mice in 3 Xi Tong Ke Li groups were longer (P<0.05).Conclusion: Xi Tong Ke Li could inhibit platelet aggregation.
