刺五加叶皂苷对大鼠心肌缺血-再灌注损伤细胞凋亡及p-ERK1/2表达的影响

摘    要:目的:研究刺五加叶皂苷(acanthopanax senticosus,ASS)对大鼠心肌缺血-再灌注损伤(myocardial ischemia reperfusion injury,MI/RI)的影响。方法选用60只SD雄性大鼠分为假手术组(C组)、缺血再灌注(ischemia reperfusion,IR)组(IR组)、刺五加叶皂苷加阻滞剂组(ASS+Z组)及刺五加叶皂苷组(ASS组),采用结扎大鼠冠状动脉左前降支建立大鼠心肌缺血再灌注损伤模型。采用NBT染色法检测心肌梗死面积及血清肌钙蛋白T(cardiac troponin T,cTnT),用TUNEL法检测心肌细胞凋亡指数(apoptotic index,AI),用Western blot法检测细胞外调解蛋白激酶(extracelluar regulated protein kinases,ERK)1/2及磷酸化ERK 1/2(p-ERK1/2)表达情况。结果 ASS组的cTnT、心肌梗死面积及AI明显低于IR组及ASS+Z组(P<0.05);ASS组p-ERK1/2表达明显高于C组、IR组及ASS+Z组(P<0.05)。结论刺五加叶皂苷对大鼠MI/RI有保护作用,能够减少心肌细胞凋亡,其机制与ERK 1/2激酶途径激活有关。

Abstract:Objective To study the effect of ASS on apoptosis and p-ERK1/2 expression in rats with myocardial ischemia-reperfusion (I/R)injury. Methods Sixty SD rats were randomly divided into sham operation group (group C), I/R group, ASS+Z group and ASS group. A rat myocardial I/R injury model was established by ligation of left anterior descending coronary artery. Myocardial infarction size was measured with nitroblue tetrazolium staining. Serum cTnT level was measured. Myocardial apoptotic index (AI) was assayed by TUNEL, and expressions of ERK1/2 and p-ERK1/2 were detected by Western blot. Results The serum cTnT level, myocardial infarction size and AI were significantly lower in ASS group than in IR group and ASS+Z group(P<0.05). The expression level of P-ERK1/2 was significantly higher in ASS group than in group C, IR group and ASS+Z group(P<0.05). Conclusion ASS can protect rats against I/R injury, attenuate myocardial apoptosis by activating the ERK1/2 signal pathway.
