摘    要:目的观察抗表皮生长因子受体(epidermalgrowthfactorreceptor,EGFR)尼妥珠单抗(h—R3)、西妥昔单抗fC225)分别联合X线照射对人鼻咽癌细胞株CNE-2的作用。方法采用流式细胞仪测定CNE-2细胞表面EGFR表达情况;采用PCR—DNA测序技术筛选K—ras野生型CNE-2细胞;采用MTS法检测细胞增殖情况;采用克隆形成实验拟合细胞存活曲线;通过流式细胞仪分析细胞周期分布及细胞凋亡变化。结果在调节CNE-2细胞克隆存活及凋亡方面,C225联合X线照射组的作用优于h—R3联合x线照射组俨〈0.05);在细胞增殖及细胞周期方面,h—R3联合X线照射组与C225联合x线照射组之间差异无明显统计学意义俨〉0.05)。结论h—R3与C225均能提高X线照射对CNE-2细胞的抗肿瘤作用,C225对该细胞株杀伤作用略优于h-R3。

Abstract:Objective To study the effect of anti-EGFR mAbs, nimotuzumab (h-R3) and cetuximab (C225), in combination with X-ray irradiation on human nasopharyngeal carcinoma cell line CNE-2. Methods Expression of surface EGFR in CNE-2 cells was detected by flow cytometry (FCM). Wild K-ras CNE-2 cells were detected by PCR-DNA sequencing. Proliferation of CNE-2 cells was assayed by MTS. Cell survival curves were plotted by clone formation experiment. Cell-cycle distribution and apoptosis were analyzed by FCM. Results The effect of combined C225 and X-ray irradiation was better than that of combined h-R3 and X-ray irradiation on the survival and apoptosis of CNE-2 cells (P 〈 0.05). However, no significant difference was observed in their effect on cell cycle distribution and proliferation (P 〉 0.05). Conclusion Both h-R3 and C225 can enhance the anti-tumor effect of X-ray irradiation on CNE-2 cells. The effect of C225 is better than that of h-R3 on CNE-2 cells.
