摘    要:目的:探讨红藻氨酸(KA)诱导大鼠癫痫发作时海马mGluR1 mRNA表达变化的时空特征.方法:采用地高辛标记的cDNA探针原位杂交法检测海马mGluR1 mRNA表达,通过图像分析系统进行定量处理.同时观察大鼠行为学及脑电变化.结果:KA注射后大鼠均出现严重边缘性惊厥,脑电图表现为持续性癫痫样放电.海马mGluR1 mRNA表达在注射KA后1h开始增高,4h ̄8h达高峰,之后逐渐回落,72h降至

Abstract:AIM: To investigate temporospatial changes of mGluR1 mRNA expression in rat hippocampus during kainic acid (KA) induced seizures. METHODS: In situ hybridization with Dig labeled cDNA probes was used to examine mGluR1 mRNA expression. The relative number of hybridization signals was quantified by image analysis systems. The EEG and behavior changes of the rats were simultaneously observed. RESULTS: All rats in the KA injected group exhibited severe limbic convulsions. The EEG recordings also showed electrical seizure activities. The mGluR1 mRNA expression began to increase 1 h after the KA injection, reached its maximal levels by 4 h~8 h, then began to decrease gradually and came to the lowest by the end of 72 h. CONCLUSION: The enhanced early expression of mGluR1 mRNA in hippocampus suggests that mGluR1 may play an important role in epilepogenesis and may be related to the subsequent neuronal damage.
