摘    要:目的 研究核苷酸切除修复基因表达水平与肺癌易感性关系,在分子水平探讨肺癌发病的相关危险因素.方法 采用病例-对照研究方法,应用实时荧光定量PCR技术检测50例原发性肺癌患者和32名健康者外周血淋巴细胞核苷酸切除修复基因(XPA、XPB、XPC、XPG、ERCC1)表达水平,分析这些基因表达水平与肺癌易感性的关系.结果 肺癌组XPB mRNA表达水平低于健康对照组(P<0.05),XPB低表达的人群患肺癌的相对危险度是高表达人群的2.57倍.结论 XPB低表达的人群肺癌发病风险增高.

Abstract:Objective To study the association between the expression levels of nucleotide excision repair genes and susceptibility to lung cancer,and explore factors related to risk of lung cancer developing in molecular level.Methods The relative expression levels of nucleotide excision repair genes(XPA,XPB,XPC,XPG,ERCC1)were measured by real time fluorescent quantitative PCR assay in a case-control study including 50 cases of lung cancer and 32 healthy controls.The association belween expression levels of five genes and susceptibility to lung cancer was analyzed.Results Expression level of XPB mRNA in lung cancer patients was reduced compared with that of controls(P<0.05).Individuals having low expression level of XPB gene exhibited a 2.5 7 fold greater risk of lung cancer.Conclusions Individuals who have low expression level of XPB gene might be at higher risk of lung cancer.
