吡喹酮对血吸虫病肝纤维化小鼠肝组织Bcl-2和Bax表达的影响

目的观察吡喹酮(Praziquantel)治疗对血吸虫病肝纤维化小鼠肝组织中与凋亡相关基因Bcl-2和Bax表达的影响。方法建立小鼠血吸虫病肝纤维化模型。模型对照组不做治疗,吡喹酮治疗组按500 mg/kg给药,1次/d,连用2 d。10周后处死小鼠,取肝组织制片,免疫组化染色(SP法),观察小鼠肝组织中Bcl-2和Bax的表达水平;HE染色观察肝组织的病理学变化。实验设正常小鼠对照。结果模型对照组和治疗组小鼠肝组织Bcl-2、Bax表达水平较正常对照组显著增高(P<0.01或P<0.05);治疗组Bcl-2水平显著高于模型对照组(P<0.05),而Bax的表达水平2组间差异无显著性(P>0.05)。结论吡喹酮可能通过促进Bcl-2表达,减少肝细胞的变性坏死,阻止血吸虫病肝纤维化的发生。

EFFECTS OF PRAZIQUANTEL ON THE EXPRESSION OF HEPATIC Bcl-2 AND Bax PROTEINS IN SCHISTOSOMIASIS MICE WITH LIVER FIBROSIS

Objective To study the effects of praziquantel on the expression of hepatic Bcl-2 and Bax proteins in schistosomiasis mice with liver fibrosis.Methods The model of schistosomiasis liver fibrosis in mice was established.The mice were divided into 3 groups: normal control group,model control group,praziquantel group.Bcl-2 and Bax proteins expression were detected by immunohistochemisty.Results Compared with normal control group,the expression of Bcl-2 and Bax was significantly higher in model control group and praziquantel group.Bcl-2 in praziquantel group was significantly higher than that in model control group,however the expression of Bax was not significantly different.Conclusion Praziquantel treatment could significantly increase expression of Bcl-2 in liver tissue of schistosomiasis mice with liver fibrosis,thereby,play its role against liver inflammation and schistosomiasis liver fibrosis.
