| 摘 要： | 目的 :建立稳定表达人胰岛素样生长因子 - 1 ( h IGF- 1 )的细胞株。方法 :用脂质体转染法将含有 h IGF- 1 c DNA的真核表达载体转染 BHK细胞 ,经 G41 8筛选 ,形成阳性细胞克隆。继续培养4周 ,进行原位杂交和免疫细胞化学检测。结果 :原位杂交检测在转染细胞的胞浆中有棕黄色颗粒样阳性杂交信号 ;免疫细胞化学检测在转染细胞的胞浆中有氯化镍蓝色阳性信号。结论 :G41 8筛选后所获得的阳性细胞克隆 ,继续培养 4周 ,原位杂交和免疫细胞化学检测表明 h IGF- 1基因得到稳定表达

| Abstract: | Objective: To establish a BHK cell line stably expressing hIGF 1. Methods: Eukaryotic expression vector plasmid containing hIGF 1 cDNA was transfected into BHK cells. The positive cells stably expressing of hIGF 1 selected by G418 was determined in situ hybridization and immunocytochemical analysis. Results: Cell clones with G 418 resistance and positive signals of situ hybridization and immunocytochemical analysis of hIGF 1 were obtained. Conclusion:The cell line that stably expresses hIGF 1 is successfully constructed in the paper. |
