| 摘 要： | 目的探讨三丁酸甘油酯(TB)对白血病细胞株HL-60细胞增殖的抑制作用,并对其作用机制进行初步探讨。方法采用MTT法观察细胞增殖变化。应用免疫组化检测抑癌基因P16的表达。应用流式细胞仪观察细胞周期的改变。通过DNA电泳观察细胞凋亡。结果TB可以抑制细胞增殖。1.0 mmol/L的TB作用72 h,有典型的DNA梯形条带。细胞周期阻滞在G0/G1期。结论TB能抑制HL-60细胞增殖,诱导细胞凋亡,其机制可能与上调P16表达有关。 |

| Abstract: | Objective To investigate the effects of tributyrin(TB) on the growth and apoptosis of HL-60 leukemia cells,and to explore its mechanism.Methods The effects of tributyrin on proliferation of HL-60 cell was determined by MTT.The P16 activity of HL-60 cells was studied by immune chemistry methods.Cell cycle was analyzed by flow cytometry.Apoptosis was analyzed by DNA ladder.Results Tributyrin inhibited the proliferation of HL-60 cells and showed in a time-dose dependent manner.HL-60 cells was treated with (1.0 mmol/)LTB for(72 h),and then the typical DNA ladder is shown on gel electrophoresis.The cells cycle of HL-60 cells was arrested in G_0/G_1.Conclusion Tributyrin could inhibit proliferation and induce apoptosis of HL-60 cells.The mechanism may relate to its up-regulation of expression of P16. |
