摘    要:目的 对从日本血吸虫尾蚴cDNA文库中筛选出的 3种新基因 ,腺苷酸激酶 (AK)、蛋白酶体激活因子PA2 8β亚单位 (PA2 8)、8kDa钙结合蛋白 (CBP)基因进行表达特性研究。方法 分别提取雌雄成虫、尾蚴、断尾童虫的总RNA ,利用RT-PCR酶混合物试剂盒 ,一步法对总RNA进行反转录及PCR扩增。结果 AK及PA2 8蛋白基因可以在雌雄成虫、尾蚴、断尾童虫体内表达 ,CBP只在尾蚴及断尾童虫中表达。结论 所筛到的AK、PA2 8、CBP基因均是日本血吸虫特异基因 ,AK及PA2 8基因可以在雌雄成虫、尾蚴、断尾童虫体内表达 ,CBP可能是日本血吸虫尾蚴、童虫的期特异性基因

Abstract:Aim To study the expression character of the three novel genes,Adenylate kinase (AK),Calcium binding protein (CBP) and Proteasome activator PA28 subunit (PA28) cDNA from Schistosoma japonicum cercariae cDNA library Method The total RNAs were extracted from female and male adult worms,cercariae and Schistosomula respectly Reverse translating and PCR were finished by one step with RT and PCR mix enzyme Result After RT PCR,AK and PA28 genes were amplified from female and male adult worms,cercariae and schistosomula total RNA,while CBP gene was only amplified from the total RNA of cercariae and schistosomula Conclusion The novel genes,AK,PA28,and CBP are specific genes of Schistosoma japonicum AK and PA28 gene are the house keeping genes of Schistosoma japonicum expressing in adult worms,cercariae and schistosomula,while CBP is only expressed in the cercariae and schistosomula stage
