原儿茶酸调节神经干/祖细胞分化的作用研究

摘    要:目的观察原儿茶酸调节体外培养的神经干/祖细胞分化及对细胞分化显型的保护作用。方法神经干/祖细胞取自胚胎14 d小鼠海马组织,以单层贴壁方式培养。免疫荧光染色标记分化的细胞,CCK-8试剂盒检测细胞的生存力,流式细胞仪分析凋亡细胞。结果原儿茶酸单独使用不能诱导神经干/祖细胞的分化,但与1%胎牛血清的联合应用时,原儿茶酸(0.06 mmol.L-1)有效地提高神经元的分化比例,介导神经元的成熟并促进突触的伸长;另一方面,原儿茶酸明显提高了分化显型的生存力并抑制了细胞凋亡,同时激活了细胞内源性的抗氧化酶系统。结论原儿茶酸能够有效地促进神经干/祖细胞分化为神经元的比例并提高细胞分化显型的生存力。

Abstract:Aim To investigate the protective effects of protocatechuic acid(PCA) on cultured neural stem/progenitor cells(NS/PCs) differentiation.Methods NS/PCs obtained from 14-day-old rat embryos were cultured under differentiation condition in the monolayer manner with or without PCA for the indicated time.Phenotypes differentiated from NS/PCs were visualized by immunofluorescent staining,the cell viability was determined by Cell Counting Kit-8(CCK-8) test,and cell apoptosis was assayed by flow cytometric analysis.Results PCA could promote the neuronal differentiation combined with fetal bovine serum(FBS) in vitro,although it could not initiate the differentiation of NS/PCs by itself.Moreover,PCA was able to induce neuronal maturation and efficiently promote neurite outgrowth.On the other hand,PCA facilitated the survival of phenotypes differentiated from cultured NS/PCs,which was associated with an increased percentage of the cellular viability and a decreased percentage of cells undergoing apoptosis under differentiation conditions.In addition,PCA-induced survival was also mediated with the activating of endogenous antioxidant enzymes.Conclusion PCA promotes the neuronal differentiation and facilitates the survival of phenotypes differentiated from cultured NS/PCs.
