中毒家兔体内盐酸哌替啶的分布研究

摘    要:目的建立LDHW-FFZ1-WuYH层析柱提取盐酸哌替啶的方法,探讨中毒家兔体内盐酸哌替啶分布规律及在氢氯噻嗪的作用下盐酸哌替啶在中毒家兔体内的分布情况。方法选择家兔6只,随机分为实验组和干扰组。实验组经耳缘静脉注射盐酸哌替啶生理盐水溶液,干扰组同样经耳缘静脉注射盐酸哌替啶生理盐水溶液,并采用氢氯噻嗪生理盐水经口灌胃,观察两组家兔的呼吸与心跳等生命体征。处死家兔后解剖,取心脏、肝脏、脾、肺、肾脏、脑组织冷冻保存待检。精确称取检材,采用LDHW-FFZ1-WuYH层析柱进行前处理,并采用气相色谱(GC/NPD)定量检测其中盐酸哌替啶水平。结果LDHW-FFZ1-WuYH提取回收率高,盐酸哌替啶平均回收率96.0%,检出限为2μg/g。实验组各脏器组织盐酸哌替啶水平由高到低分别为脑组织(3.45±0.31)μg/g]、肾脏(2.65±0.16)μg/g]、肝脏(2.15±0.12)μg/g]、脾(1.81±0.13)μg/g]、肺(1.35±0.23)μg/g]、心脏(1.13±0.21)μg/g]。干扰组各脏器组织盐酸哌替啶水平由高到低分别为脑组织(1.75±0.14)μg/g]、肾脏(1.45±0.42)μg/g]、肝脏(1.19±0.24)μg/g]、脾(0.98±0.26)μg/g]、肺(0.75±0.26)μg/g]、心脏(0.51±0.41)μg/g]。干扰组心脏、肝脏、脾、肺、肾脏、脑组织标本盐酸哌替啶水平均低于实验组,差异有统计学意义(P<0.05)。结论建立了LDHW-FFZ1-WuYH层析柱提取盐酸哌替啶的方法,该方法可用于盐酸哌替啶中毒致死案件检材的前处理。氢氯噻嗪可加快家兔体内盐酸哌替啶的排泄。

Abstract:Objective To establish a method for extracting meperidine from LDHW-FFZ1-WuYH column and to study the distribution of meperidine of postmortem in poisoned rabbits and the distribution of meperidine poisoned rabbits under the action of hydrochlorothiazide.Methods Six rabbits were randomly divided into experimental group and control group.Experimental group rabbits were injected meperidine saline solution through ear vein,and the control group were also injected meperidine saline solution through ear vein,as well as hydrochlorothiazide saline solution by gavage.The respiratory and heartbeat were observed.In addition,postmortem anatomy,coring,liver,spleen,lung,kidney and brain cryopreservation were examined.Using LDHW-FFZ1-WuYH column,sample was handled,and gas chromatography(GC/NPD)was used to detected meperidine.Results LDHW-FFZ1-WuYH had a higher recovery rate,with an average recovery rate of 96.0%and the detection limit was 2μg/g.In experimental group,the levels of meperidine from high to low in various organs were brain(3.45±0.31)μg/g],kidney(2.65±0.16)μg/g],liver(2.15±0.12)μg/g],splenic(1.81±0.13)μg/g],lung(1.35±0.23)μg/g]and heart(1.13±0.21)μg/g].In control group,the levels of meperidine from high to low in various organs were brain(1.75±0.14)μg/g],kidney(1.45±0.42)μg/g],liver(1.19±0.24)μg/g],spleen(0.98±0.26)μg/g],lung(0.75±0.26)μg/g],heart(0.51±0.41)μg/g].The levels of meperidine of heart,liver,spleen,lung,kidney and brain tissue in control group were significantly lower than that of experimental group.Conclusion A method of extracting meperidine from LDHW-FFZ1-WuYH lamination column is established.It can be applied to the pretreatment of meperidine′s lethal case.The use of hydrochlorothiazide could accelerate the excretion of meperidine.
