摘    要:目的调查湖北土家族人群24个常染色体短串联重复(STR)基因座遗传多态性,探讨其群体遗传关系及在法医学中的应用价值。方法应用SureID? PanGlobal对457例湖北土家族无血缘关系个体的DNA进行扩增,3500XL型遗传分析仪进行电泳分析,GeneMapper ID-X v1.5软件分析等位基因片段大小。统计分析24个STR基因座的频率数据和法医遗传学参数,并与其他地区人群已有数据进行比较。结果湖北土家族人群各基因座个体识别概率(DP)为0.797 2~0.991 7,多态性信息含量(PIC)为0.561 5~0.939 1。累积个体识别率(CDP)和累积非父排除率(CPE)分别为1-2.574 0×10-30和1-1.593 9×10-11。从Nei′s DA遗传距离矩阵分析发现,湖北土家族与广东汉族的遗传距离最小(0.033 5),与云南苗族遗传距离最大(0.061 6)。结论 24个STR基因座在湖北土家族人群中具有丰富的遗传多态性。研究不同民族群体的遗传多样性对了解他们的起源、迁移以及相互关系有重要的意义。

Abstract:Objective To investigate the genetic polymorphisms of 24 autosomal short tandem repeats(STR)loci in Hubei Tujia population,and to explore the population genetic relationships and their application value on forensic medicine.Methods The DNA samples of 457 unrelated individuals of Hubei Tujia population were amplified using SureID PanGlobal kit,and the PCR products were analyzed by electrophoresis through 3500XL genetic analyzer.The fragment sizes of alleles were analyzed subsequently by GeneMapper ID-X v1.5.Allele frequencies and forensic genetic parameters of 24 STR loci were statistically analyzed and compared with the available data of other populations from different races and regions.Results The discrimination power(DP)were 0.7972 to 0.9917,and polymorphic information content(PIC)were 0.5615 to 0.9391.The total discrimination power(CDP)and cumulate probability of exclusion(CPE)were 1-2.5740×10-30 and 1-1.5939×10-11,respectively.Compared with 24 reference populations,the genetic distance(Nei′s DA)between Hubei Tujia population and Guangdong Han population was the smallest(0.0335),while it was largest between Hubei Tujia population and Yunnan Miao population(0.0616).Conclusion The 24 STR in Hubei Tujia population has abundant genetic polymorphism.The study of genetic diversity among different populations is useful in research of their origins,migrations and their relationships.
