亚慢性镉中毒所致小鼠性腺损伤及海尔福保护作用研究

摘    要:目的了解亚慢性镉中毒所致小鼠性腺损伤及海尔福保护作用。方法按雌雄各半随机将动物分为三组,染毒剂量为1.0mg/kg腹腔注射染毒,每日一次,每周5d,连续7周,建立亚慢性镉中毒模型。治疗组腹腔注射氯化镉溶液的同时给予1.5nW只海尔福溶液灌胃。实验设立对照组(生理盐水腹腔注射)。染毒20d起连续观测并记录动情周期。实验结束后,观察各组睾丸、卵巢组织病理改变,测定卵巢、睾丸镉含量。结果中毒组睾丸组织病理改变明显,卵巢组织病理改变不明显,但闭锁卵泡构成比显著高于对照组,卵巢、睾丸镉含量明显高于对照组,动情周期与对照组比明显延长;治疗组睾丸组织损伤减轻,闭锁卵泡构成比与对照组无明显差异,卵巢、睾丸镉含量明显低于中毒组,动情周期与对照组比无明显差异。结论镉对性腺有明显的毒性作用,海尔福对镉的生殖毒性有一定的保护作用。

Abstract:Objective To explore the subchronic of toxicity of cadmium on sexsual glands and the protective effect of hai-er-fu in mice model. Method Mice were divided into 3 groups. Two experimental groups were injected subcutaneously with 1.0mg/kg of cadmium chloride(Cdcl_2 ) for 7-week (once per day,five days per week), one groups received hai-er-fu 1.5ml in drinking water; and other group fed on distilled water served as blank control group. At the end of the 20-day experimental period, the estrus cycle of mice was monitored, pathologic changes of testis and ovary were examined and the cadmium content measured. Results The cadmium content in the ovary and testis was significantly increased and the estrous cycle was delayed significantly in exposed mice than that in the control mice. Pathologic changes revealed obvious testicular tissue damaged, and the ovarian tissue only slightly damaged. The proportion of atresic follicle was significantly increased in the exposed than that of treated groups. Hai-er-fu ameliorates significantly the pathologic changes of testis, and the proportion of atresic follicle and the estrous cycle in treated groups was similar as compared with the control group. The cadmium content in the ovary and testis was significantly decreased in the treatment group. Conclusion Cadmium shows subchronic toxicity on both male and female sexual glands,and Hai-er-fu demonstrates protective effect against cadmium toxicity to a certain extent.
