摘    要:目的:评价白藜芦醇(Resveratrol,Res)对皮肤鳞状细胞癌(简称鳞癌)移植瘤生长的影响。方法:建立人皮肤鳞癌(A431)裸鼠移植瘤模型,分别予以药物干预后处死荷瘤鼠,称取瘤质量;HE染色观察各组移植瘤组织细胞病理形态学变化;应用原位末端标记(TUNEL)技术检测细胞凋亡;采用sP免疫组化法检测Res对各组裸鼠移植瘤组织中凋亡调控基因Bcl-2及Bax的蛋白表达的影响。结果:Res实验组移植瘤体积、瘤质量低于阴性对照组,肿瘤组织的凋亡指数高于阴性对照组,瘤组织中Bcl-2蛋白表达低于阴性对照组,Bax蛋白表达高于阴性对照组(均P〈0.05)。结论:白藜芦醇可通过下调Bcl-2和上调Bax蛋白的表达诱导皮肤鳞癌细胞凋亡,从而抑制移植瘤的生长。

The impact of resveratrol on human skin squamous cell carcinoma A431 xenografts growth in nude mice

Abstract:Objective: To evaluate of the impact of resveratrol (Res) on human skin squamous cell carcinoma A431 xenografts growth in nude mice. Methods: The human skin squamous cell carcinoma (A431 ) xenograft model in nude mice was established and randomly divided into the blank group, the Res groups and the saline negative con- trol group, respectively. After intervention with Res, the mice were killed. The tumor weight and volume were recorded. The pathological change of tumor tissue in each group was observed under microscope with HE staining. The tumor apoptosis was detected by TUNEL technique. The impact of Res on the protein expression of apoptosis regulatory genes, Bcl- 2 and Bax, was detected by SP immunohistochemistry assay. Results: The tumor volume and weight in Res groups were less than those in the negative control group ( P 〈 0.05). The apoptotic index of the tumor tissue was higher in Res groups than in the negative control group ( P 〈 0.05). The protein expression of Bcl - 2 was lower in Res groups than in the control group, while the protein expression of Bax was higher in Res groups than in the control group (P 〈 0.05). Conclusion: Res can inhibit the growth of human skin squamous cell carci- noma A431 xenograft in nude mice through down - regulating the Bel - 2 protein expression and up - regulating the Bax protein expression, resulting in apoptosis of the tumor cells.
