| 摘 要： | 目的 重组腺病毒介导HOSM基因转染胰腺癌细胞后，用RT-PCR方法检测外源性HOSM基因的表达情况。方法 构建了含人抑瘤素M(OSM，oncostatin M)基因的复制缺陷型重组腺病毒载体ad-HSOM，以ad-HOSM转染胰腺癌细胞株后，提取细胞的总RNA，用RT-PCR法检测HOSM基因在胰腺癌中的表达。结果 HOSM基因在转染细胞中有效的表达。结论 RT-PCR法能有效的检测外源性HOSM基因在胰腺癌中的表达，为重组腺病毒介导HOSM基因用于胰腺癌治疗的实验研究奠定了基础。

| Abstract: | Objective RT PCR assay was used to identify the expression of exogenous HOSM gene in pancreatic carcer cell line which is transfected by HOSM gene with recombinant adenovirus.Methods To construct a replication defective recombinant adenoviral vector encoding HOSM gene(ad hosm),to extract total RNA from the ad HOSM transfected cells,RT PCR assay was used to identify the expression of exogenous HOSM gene in a pancreatic carcer cell line.Results The exogenous HOSM gene was espressed effectively in the transfected pancreatic cell.Conclusion RT PCR assay was able to detect the expression of exogenous HOSM gene in pancreatic carcer cells,and to settle the foundation for found the experimental study of the treatment of pancreatic neoplasm by HOSM gene via recombinant adenovius. |
