摘    要:从柔红霉素产生菌SIPI-1482菌株基因组DNA中PCR扩增得dnmV及其上游dnmU基因片段。利用同源重组对SIPI-1482菌株的dnmV基因进行基因阻断,得到一株遗传稳定的破坏于。验证了在该菌株中进行同源重组所需交换臂长度。

Abstract:dnmV was PCR amplified along with its upstream dnmU gene from genomic DNA of Streptomyces coeruleorubidus SIPI-1482, which was one of the daunorubicin-producers. Homologous recombination was used for dnmV gene disruption in SIPI-1482, and one genetically stable disruptant was obtained. The homologous DNA fragment which was sufficient in length for recombination to occur in this strain was identified.
