大鼠急性出血坏死性胰腺炎的内毒素血症与组织改变的关系及纳屈酮治疗作用

摘    要:目的:应用牛磺胆酸钠诱发大鼠急性出血坏死性胰腺炎(AHNP)模型,探讨大鼠AHNP的内毒素血症与胰腺组织改变的关系及纳屈酮治疗作用。方法:实验分为三组:对照组,AHNP组及纳屈酮(NTX)治疗组;以6、12、24为观测时点,观察三组大鼠血浆淀粉酶、内毒素、胰腺系数及胰腺组织变化。结果:发现对应各时点AHNP组较对照组的血浆淀粉酶、内毒素(ET)及胰腺系数升高,胰腺组织明显损害;纳屈酮(NTX)治疗组各时点较AHNP组的血浆淀粉酶、ET及胰腺系数下降;而且NTX组胰腺损害减轻。结论:NTX可能通过降低血浆ET,而减轻胰腺损害,延长AHNP大鼠生存时间及降低病死率。

Abstract:Objective: A model of acute hemorrhagic necrotizing pancreatitis(AHNP) was induced by retrograde injection of 5% sodium taurocholate into the pancreatic duct for exploring the relationship between endotoxemia and histologic changes in the pancreatic tissue and therapeutic effects of naltrexone on AHNP in rats. Methods: The rats were randomly divided into three groups: control group, AHNP group and NTX treatment group. We studied the effects of morphine receptor blocker, naltrexone during the rats of AHNP and measured the plasma levels of amylase, pancreatic index, endotoxin and calculated the death rate as well as average survival time in rats of both AHNP and NTX groups. Changes of the pancreatic tissue was examined by light and electric microscopy. Results: The results demonstrated that amylase, endotoxin were significant fall in the plasma; besides, the damage of pancreatic tissue was reduced. Conclusion: NTX decreased endotoxin in the plasma and improves the damage of pancreatic tissue. The leth ity rate in rats of AHNP was significantly lower in NTX treatment.
