摘    要:目的:探讨积雪草酸(AA)对小鼠心肌组织缺血损伤的保护作用及其可能机制。方法:通过开胸结扎左前降支建立小鼠心肌梗死模型。实验分为3组:假手术组(A组);手术组(B组);药物干预组(C组)。术后两周,心脏超声检测心功能;EVANS蓝-TTC联合染色检测心肌梗死面积;Hoechst染色法测定各组心肌细胞凋亡率;Western blot法检测心肌组织中磷酸化-p38、p38蛋白的表达;荧光显微镜检测DHE荧光强度。结果:B、C组心肌细胞凋亡率、磷酸化p38表达、DHE荧光强度高于A组(P<0.05),而C组上述指标低于B组(P<0.05)。结论:积雪草酸对小鼠心肌梗死后的心脏功能具有一定的保护作用,其机制可能与降低磷酸化p38蛋白的表达水平有关。

Abstract:Objective: To explore the protective effect and mechanisms of asiatic acid (AA) on myocar-dial ischemia injury (MI) in mice. Methods: Myocardial infarction models were established by ligating the left anterior descending coronary artery. Thirty C57BL/6 mice were divided into three groups of A (Sham operation group), B (myocardial infarction group, after operation, the mice were treated with vehicle for 2 weeks) and C (drug intervention group, after operation, the mice were treated with AA for 2 weeks). Two weeks after surgery, cardiac function were examined by two-dimensional echocardiography, myocardial infarct size was detected by Evans blue-TTC staining. Cell apoptosis was confirmed by Hoechst method, p-p38 and p38 protein expression were determined by Western blot. Results: Cardiac function, Cardiac cell apoptosis rate, infarct size and ex-pression of p-p38 in group B and C were higher than those in group A (P<0.05), which were lower in group C than those in group B (P<0.05). Conclusion: AA can improve the heart function in mice with myocardial infarctions. The protective mechanism may be the inhibition of p-p38 expression.
