摘    要:目的建立测定犬脑组织中异丙酚含量的高效液相色谱方法。方法采用ZORBAXEclipseXDB-C18(4.6×150mm,5μm)为色谱柱,以甲醇-水(80:20,v/v)为流动相,流速1.0ml/min,检测波长为270nm。结果异丙酚的线性范围0.1~8μg/ml(r=0.9998),最低检测限和最低定量限为0.02μg/ml(S/N=3)和0.08μg/ml(S/N=10)。低、中、高浓度的平均回收率分别是89.5%、92.0%、96.16%,日内精密度和日间精密度分别为5.0%、4.2%、4.55%和6.74%、2.72%、5.07%。结论本方法快速、简单、准确,可用于测定犬脑组织和血浆中异丙酚的含量。

Abstract:Objective To develop an HPLC-UV method for determination of propofol in dog brain tissue. Methods Chromatography separation was performed on a ZORBAX Eclipse XDB-C18 (4. 6 × 150mm, 5μm) with the mobile phase consisted of methanol water (80/20, V/V) at room temperature. The flow rate was 1 ml/min and the monitor wave- length was 270nm. Results The linear range for propofol was 0. 1-8μg/ml with r=0. 9998. The LOD and LOQ were 0.02vg/ml(S/N= 3) and 0. 08μg/ml (S/N= 10), respectively. The average recoveries of low, medium and high concentration were 89.5%, 92.0% and 96.16 %, respectively. The RSD% of intra-day (n= 5) and inter-day (n= 3) was below 7%. Conclusions The method was sensitive, accurate, precise and reliable for the determination of propofol in dog brain tissue.
