肾衰泄浊颗粒主要药效学及急性毒性实验研究

目的:观察肾衰泄浊颗粒对慢性肾功能衰竭(CRF)大鼠的影响并观察其急性毒性反应。方法:建立CRF大鼠模型,观察CRF大鼠使用肾衰泄浊颗粒前后肾功能及血液学指标的变化情况。将肾衰泄浊颗粒以最大浓度给小鼠灌胃,小鼠没有出现死亡,未测出LD50故测其最大耐受量。结果:肾衰泄浊颗粒能明显降低腺嘌呤所致CRF大鼠的血肌酐(Scr)、尿素氮(BUN)水平,能提高红细胞(RBC)、血红蛋白(HGB)和红细胞压积(HCT)。肾衰泄浊颗粒给小鼠灌服的最大耐受量可达46.16g/kg,相当于临床用量的276.96倍。结论:肾衰泄浊颗粒是治疗CRF大鼠的有效药物,且具有较好的安全性。

Study of the Shenshuai Xiezhuo GranuIe in Main Pharmacodynamics and Acute Toxicity

Objective:To observe the effect of Shenshua Xiezhuo(SSXZ)granule on chronic renal fai lure(CRF)and acute toxicity of SSXZ granule.Methods:To construct CRF rat model,and to observe the changes of renal function and blood routine of CRF rat.Intragastric administration was taken for mice with maximum concentration SSXZ,but no mice die.Median lethal dose(LD50)was not measured.So maximally tolerated dose was measured.Results:SSXZ granule can markedly cut down the levels of serum blood creatinine(Scr),urea nitrogen(BUN)of CRF rat,and it can markedly ameiiorate the levels of red blood cells(RBC),hemoglobin(HGB)and hematocrit(HCT).The maximally tolerated dose was 46.16g/kg in mice for SSXZ granule,which is 276.96 times of clinical dose.Conclusion:SSXZ granule is a effective medicine for CRF rat and safe.
