三生饮对大鼠局灶性脑缺血Bcl-2 Bax蛋白表达的影响

摘    要:目的:探讨三生饮对大鼠局灶性脑缺血细胞凋亡相关基因Bcl-2和Bax蛋白表达影响。方法:建立大鼠局灶性脑缺血再灌注模型,采用TTC梗死面积测定及SP免疫组织化学方法,检测再灌注后3h、6h、12h、2 4h、3d大鼠大脑皮质的Bcl-2、Bax免疫反应阳性细胞平均计数,比较模型组与三生饮治疗组效果。结果:与模型组相比三生饮可减轻脑缺血程度,减小梗死体积(P <0 0 1) ;促进Bcl -2蛋白表达,延长表达时程(P <0 0 1) ;而对Bax蛋白的表达则无抑制作用,两组间无显著性差异(P >0 0 5 )。结论:三生饮可能通过提高抗凋亡基因表达,从而对脑缺血再灌注损伤起到一定的保护作用

Abstract:Objective :To study the effect of Sanshengyin on the expression of Bcl 2 and Bax protein in the rats after focal cerebral ischemia. Methods :Creation the rat model of middle cerebral artery occlusion (MCAO) and reperfusion.Using the tetraphenyl tetrazolium chloride(TTC) staining detected the ischemic zone,Immunohistochemistry method analyze the expression of Bcl 2 and Bax protein positive cells at 3h,6h,12h,24h,3d after reperfusion.Compared the results between the group of MCAO and Sanshengyin. Results :Compared with the results of the MCAO group,Sanshengyin could relieved the lesion of cerebral ischemia;reduced the size of the infarct;and in this group,Bcl 2 protein positive cells increased obviously.But Bax protein positive cells did not decrease,the results of the two group have no significant differentiation( P >0 05). Conclusion :Sanshengyin is the effective neuroprotective by increasing the expression of Bcl 2 protein,when the cerebral ischemia reperfusion injury occurring.
