摘    要:目的:观察固本、宣肺、宣肺固本3种中医治法对哮喘大鼠气道重建的作用及机制.方法:利用卵蛋白复制哮喘大鼠模型,予以灌胃给药治疗,分别利用嗜酸性粒细胞计数液和ELISA方法检测大鼠肺泡灌洗液(BALF)中嗜酸性粒细胞(EOS)数和血清中基质金属蛋白酶-9(MMP-9)及其抑制物-1(TIMP-1)的表达.结果:经过固本、宣肺、宣肺固本3种治法中药治疗,哮喘大鼠BALF中EOS计数均降低,血清中MMP-9的表达水平降低、TIMP-1的表达升高,与模型组均有统计学差异.结论:固本、宣肺、宣肺固本3种治法对哮喘大鼠炎症及气道重建均具有改善作用,其作用机制可能与减少嗜酸性粒细胞肺泡浸润,下调MMP-9表达与上调TIMP-1表达有关.

Abstract:This paper was aimed to observe protective effects of traditional Chinese medicine (TCM) varying in therapeutic methods on airway remodeling in bronchial asthma. Three TCM decoctions, representing body resistance-strengthening therapy, lung-diffusing therapy, and combination therapy were prepared respectively and administrated peroral to asthmatic rats induced by ovalbumin. Eosinophils (EOS) were counted in bronchial veolarlavage fluid (BALF), and matrix metalloproteinase (MMP-9) and tissue inhibitor of metalloproteinase (TIMP)-1 levels in serum were detected by ELISA. The results showed that compared with the control group, administration of TCM recipes is significantly effective in reducing EOS infiltration into BALF, as well as drop in MMP-1 level whereas rise in TIMP-1 level. It was concluded that three TCM therapies exerted similar protective effects against bronchial asthma. The improvement in airway remodeling can be attributed in part to inhibiting infiltration of EOS, down-regulating MMP-9 expression and up-regulating TIMP-1 expression.
