rhBMP2缓释微球对牙龈成纤维细胞的生物学作用

摘    要:目的:观察甲基丙烯酸缩水甘油酯右旋糖酐(dex-GMA)载重组人骨形态发生蛋白-2(rhBMP2)缓释微球(MPs)对体外培养的人牙龈成纤维细胞(GFCs)的生物学作用。方法:采用体外细胞培养技术,将rhBMP2缓释微球和GFCs一起培养,采用M1Tr法检测其对GFCs的增殖状况的影响;碱性磷酸酶(ALP)检测试剂盒检测细胞ALP活性以反映rhBME2缓释微球对其分化状况的影响,并与单纯rhBMP2的作用进行比较。结果:rhBMP2缓释微球具有良好的生物学活性,能显著促进的GFCs增殖及分化,并维持10d以上,其效应高于单纯施加rhBMP2的效应。结论:所制备的rhBMP2微球比单纯rhBMP2对GFCs有更为明显的生物学效应,可以达到对生长因子的有效缓释。

Biological Effects of Recombinant Human Bone Morphogenetic Protein-2 Loaded Dextran-based Microspheres on Gingival Fibroblast Cells in Vitro
Abstract:Objective: To evaluate the biological effects of recombinant human bone morphogenetic protein-2 (rhBMP_2) loaded dextran-based microspheres (rhBMP_2-dex-MPs) on cultured gingival fibroblast cells (GFCs) in vitro. Methods: Cell culture technique and MTT colorimetric assay were used to evaluate the proliferation of the GFCs, ALP kit was used to evaluate the ALP activity of the GFCs so as to show the differentiation of the cells by adding the rhBMP_2-dex-MPs or rhBMP_2 only. Results: The results showed that rhBMP_2-dex-MPs had biological activity, and statistical analysis showed that rhBMP_2-dex-MPs could enhance significantly both proliferation and differentiation of human GFCs more than 10 days, the effect of the rhBMP_2-dex-MPs was significantly higher than that of rhBMP_2. Conclusion: The biological effects of rhBMP_2-dex-MPs were significantly higher on cultured cells than that of rhBMP_2 only. Encapsulating rhBMP_2 into dex-MPs may be an effective way of rhBMP_2 sustained release in tissue engineering work.
