热疗联合紫杉醇对NCI-H446细胞增殖和VEGF mRNA表达的影响

摘    要:目的:观察热疗联合紫杉醇对NCI-H446细胞增殖作用以及VEGF mRNA表达的影响。方法:将NCI-H446细胞分组,Ⅰ热疗组(43℃加热90 min);Ⅱ紫杉醇组;Ⅲ热疗联合紫杉醇组;Ⅳ对照组(常规培养细胞)。分别在处理72 h后用MTT法检测4组细胞增殖情况;RT-PCR检测各组细胞中VEGF mRNA的表达量。结果:加热抑制了NCI-H446细胞的增殖;单独应用紫杉醇随着药物浓度的倍比增大,增殖抑制率逐渐增大;加热联合紫杉醇与对应浓度单独使用比较差异有统计学意义(P<0.05)。加热降低了NCI-H446细胞中VEGF mRNA的表达;加热联合紫杉醇组中NCI-H446细胞中VEGF mRNA的表达量明显减少,与对应浓度紫杉醇组比较差异有统计学意义(P<0.05)。结论:43℃加热90 min联合紫杉醇对NCI-H446细胞增殖有抑制作用,并降低了VEGF mRNA的表达量。

Abstract:Objective: To observe the effects of hyperthermia combined with paclitaxel on NCI-H446 cells proliferation and VEGF mRNA expression.Methods: NCI-H446 cells were divided into 4 groups:Hyperthermia at 43 ℃ for 90 min were given to group Ⅰ.NCI-H446 were cultured with different paclitaxel in group Ⅱ.NCI-H446 were treated with hyperthermia combined with paclitaxel in group Ⅲ,while normally cultured NCI-H446 were as control(group Ⅳ).MMT assay was adopted to measure cell proliferation and the inhibition rates.Semi-quantitive RT-PCR was applied to determine the expression of VEGF mRNA.Results: both Thermotherapy and paclitaxel could inhibit the proliferation of NCI-H446 cells and thermotherapy enhanced the inhibitive effects of paclitaxel on the NCI-H446.Thermotherapy and thermochemotherapy could decrease the expression of VEGF mRNA.Conclusion: Hyperthermia at 43 ℃ for 90 min combined with paclitaxel could inhibit cell proliferation and decrease the expression of VEGF mRNA on NCI-H446 cells.
